CP110 Suppresses Primary Cilia Formation through Its Interaction with CEP290, a Protein Deficient in Human Ciliary Disease

Primary cilia are nonmotile sensory organelles implicated in cellular signaling. The centrosomal protein CP110 inhibits primary cilia formation through an unknown mechanism. Tsang et al. show that CP110 interacts with CEP290, a protein deficient in human ciliary disease. CP110 antagonizes CEP290 function, which cooperates with Rab8a to promote ciliogenesis.

Issue Highlights

Molecular mechanisms that link organ patterning to organ growth remain mysterious. Rogulja et al. show that in the Drosophila wing, a BMP morphogen gradient controls graded expression of Fat/Hippo signaling pathway regulators. They present evidence that patterning is coupled to growth control via this mechanism.
Endocytosis of cell surface signaling receptors can either prolong signaling activity through recycling or attenuate it through receptor degradation. Here, Sigismund et al. show that, contrary to previous expectations, in the case of the EGFR, clathrin-mediated internalization promotes recycling and prolongs the signal duration, whereas clathrin-independent endocytosis leads to degradation.
Tight control of transposons is essential for the integrity of the germline. Soper et al. demonstrate that Maelstrom, a component of a germ cell-specific organelle called nuage, is indispensable for DNA methylation-controlled silencing of transposons in meiosis and for homologous chromosome synapsis in the mouse male germline.

In humans, mutations in genes encoding Sonic hedgehog (Shh) and the homeobox protein SIX3 cause holoprosencephaly, a common forebrain malformation characterized by incomplete separation of the cerebral hemispheres. Geng et al. reveal that Six3 proteins carrying holoprosencephaly-associated mutations have decreased activity and fail to activate Shh expression, thereby causing holoprosencephaly.
