Magnification of the Leading Edge Region at the Beginning of Dorsal Closure
from an Embryo Expressing α-Catenin-GFP in Lateral Epidermal Tissue
and in the Larger Amnioserosa Cells
The movie covers the "initiation" and "sweeping" phases of dorsal closure. As the lateral epithelial cells elongate and move forward, the amnioserosa cells reduce their apical surface and the leading edge of the front row cells contracts and becomes taut.
Movie S2. Final Stages of Dorsal Closure, Including the "Zippering" and "Termination" Stages, Were Revealed in Another α -Catenin-GFP Embryo
The opposing epithelial fronts zipper together from both anterior and posterior ends of the dorsal hole and form new adhesions in the midline. During this process, the apical surface of the amnioserosa contracts and disappears beneath the closing epithelia.

The leading edge cells express a thick taut actin cable and extensive filopodial protrusions. As the epithelia move forward the two epithelial fronts get closer and filopodia from opposite sides reach one another and draw the two fronts together.
